1. Begin PCR White Boarding Activity

            • Start with one dbl-stranded segment of DNA

            • End with dbl-stranded short product at the beginning of the 4th cycle

            • Never draw an actual helix – rather draw linear two-sided DNA

            • Label and color-code primers, template DNA, synthesized DNA, Taq,
               primer target sequence, 5 & 3 ends

            • Original primers must be interior to the ends of the
               original short-product template

            • in preparation for the 2 labs after the Lambda Amplification Lab

            • prepping and running gels, and into the staining tray

1. Lambda Amplification Lab

            • Day 1: PCR Reaction, and Amplification,
              using Lambda genome as the template

            • PCR product remains in the thermal cycler at 15˚C

            ramping video

            • Day 2: gel electrophoresis of PCR product,
              gel staining, and gel photographing
